Experiment: Carbon Dioxide Fixation in Intact Cells

In this month's experiment, the amount of carbon dioxide consumed during photosynthesis is measured in the presence and absence of light and chemical reagents.  The reactions that fix carbon dioxide into sugars take place during the portion of photosynthetic process known as the Calvin Cycle.  The chemical agents used in this experiment affect either the light-driven reactions that produce high-energy molecules used in the Calvin Cycle, or the enzymes involved in assembly of the sugar molecules during the cycle. 

The consumption of carbon dioxide by intact cells, during photosynthesis, is quantified by measuring the production of oxygen with a polarograph.  Since one molecule of oxygen is produced for every molecule of carbon dioxide consumed, it is easy to determine the amount of carbon dioxide consumed by using an ISE-730 dissolved oxygen electrode, a DO2-100 current to voltage adapter that converts the output of the ISE-730 to measure any changes in oxygen concentration of the chamber.

Tech Notes: The RPC-100 Respiration/Photosynthesis Chamber, the ISE-730 Oxygen Electrode, and the DO2-100 Current to Voltage Adapter

The RPC-100 is a reaction chamber designed for studying cellular respiration or photosynthesis.  The chamber is filled with a mixture of cells or cell fractions, buffers, and reagents that react to either consume or produce oxygen.  The amounts and rates of oxygen, either consumed or produced, are measured using an ISE-730 oxygen electrode placed in the chamber.  A DO2 adapter that also amplifies the output of the electrode converts the current output of the electrode to a voltage.
